Mechanism of action

The Pedunculopontine Nucleus (PPN) is the fall prevention center of the brain. It controls gait, balance, and cognition. Nicotinic receptors are distributed throughout the central nervous system with clusters in the PPN. NP002, a proprietary formulation of nicotine, is the archetypal nicotinic acetylcholine agonist.

Degeneration of the PPN causes a loss of acetylcholine signaling in the basal ganglia/striatum, resulting in an increase in falls. NP002 directly stimulates nicotinic acetylcholine receptors, which replaces the missing acetylcholine from lost PPN neurons and thereby decreases falls.
